Retatrutide: from the 2023 Phase 2 trial to 2026 Phase 3 presentations
The triple agonist (GIP, GLP-1, glucagon) now has Phase 3 obesity data presented in 2026. It is still not an approved drug.
Retatrutide is a single peptide engineered to activate GIP, GLP-1 and glucagon receptors. The 2023 Phase 2 *NEJM* trial reported large 48-week weight reductions, with dose-related gastrointestinal effects and a transient rise in heart rate.
What 2026 added
The TRIUMPH Phase 3 program began reporting in 2026, including a June 2026 ADA presentation of TRIUMPH-1 in obesity (with subsets for sleep apnea and knee osteoarthritis). Conference slides are not a substitute for a peer-reviewed paper, but they mark the move from Phase 2 signal to Phase 3 testing.
What is still true
Retatrutide is not FDA-approved. Phase 3 safety, durability after stopping, and peer-reviewed publications will decide whether the Phase 2 magnitude holds. Research-chemical “retatrutide” sold online is not the trial product.
